VNRX-9945 is a third-generation, orally bioavailable small molecule core protein allosteric modulator (CpAM) developed for the treatment of chronic hepatitis B virus (HBV) infection. It acts as a direct-acting antiviral by inhibiting HBV replication through dual mechanisms—blocking nucleocapsid assembly and preventing the formation of covalently closed circular DNA (cccDNA). Preclinical studies have shown that VNRX-9945 exhibits potent and broad antiviral activity against multiple HBV genotypes, suppressing viral DNA to undetectable levels in animal models. The drug has demonstrated favorable pharmacokinetics supporting once-daily dosing and was well tolerated in toxicology studies. As of 2021, it entered Phase 1 clinical trials to evaluate safety and pharmacokinetics in healthy volunteers[1][2][3][4][6].
