VOB560 is a potent and selective small molecule inhibitor of B-cell lymphoma 2 (BCL2), developed by Novartis for the treatment of hematological malignancies. By binding to and inhibiting BCL2, VOB560 restores apoptotic processes in tumor cells, promoting cell death in cancers that rely on this anti-apoptotic protein for survival. It has demonstrated activity against BCL2 mutations such as G101V and D103Y. Clinical development has focused on relapsed or refractory acute myeloid leukemia (AML), non-Hodgkin lymphoma (NHL), chronic lymphocytic leukemia (CLL), and multiple myeloma (MM). In clinical trials, it is also being studied in combination with MIK665, an MCL1 inhibitor, to enhance pro-apoptotic effects[1][4][5][6][9].
