Overview

Vobarilizumab is a humanized bispecific nanobody (Llama-derived heavy-chain only Vhh antibody) designed to block the interleukin-6 receptor (IL6R). It consists of an anti-IL6R domain and an anti-human serum albumin domain, the latter extending its half-life. Vobarilizumab preferentially binds soluble IL6R over membrane-bound IL6R and inhibits signaling by interleukin 6, a pro-inflammatory cytokine involved in T-cell activation, acute phase protein production, immunoglobulin induction, and osteoclast differentiation. The drug was developed for inflammatory autoimmune diseases such as rheumatoid arthritis and systemic lupus erythematosus. It has demonstrated strong disease-modifying activity in clinical trials for rheumatoid arthritis[1][3][4][5][6].
