Overview

Voclosporin is a small molecule calcineurin inhibitor used as an immunosuppressant for the treatment of active lupus nephritis in adults with systemic lupus erythematosus. It is a structurally modified analog of cyclosporin A, designed to enhance its action against calcineurin. Voclosporin inhibits calcineurin, which blocks the production of interleukin-2 (IL-2) and T-cell mediated immune responses, leading to reduced inflammation and stabilization of podocytes in the kidneys. This dual mechanism helps prevent further renal damage by both suppressing immune activity and promoting podocyte stability[1][5][6][8]. Voclosporin was developed by Aurinia Pharmaceuticals and is marketed under the brand name Lupkynis[1][2]. It received FDA approval in January 2021 for use in combination with mycophenolate mofetil and corticosteroids for lupus nephritis[2].
