Overview

Vofatamab is a fully human immunoglobulin G1 (IgG1) monoclonal antibody that targets the fibroblast growth factor receptor 3 (FGFR3). It acts as an antagonist by binding to both wild-type and mutated forms of FGFR3, thereby inhibiting its activation. This mechanism blocks ligand-induced activation as well as signaling from mutant or fusion forms of the receptor. Vofatamab was developed primarily for the treatment of urothelial cell carcinoma and other FGFR-driven cancers. The drug was initially developed by BioClin Therapeutics and later advanced by Rainier Therapeutics. Clinical trials investigated its use in patients with metastatic urothelial cancer who had failed prior chemotherapy regimens, both as monotherapy and in combination with docetaxel. Despite early promise in clinical studies, development has been discontinued[1][2][5][6][7][8].
