**Vofatamab + actinium-225 (FPI-1966)** is a radiopharmaceutical composed of vofatamab, a human IgG1 monoclonal antibody that targets fibroblast growth factor receptor 3 (FGFR3), conjugated via the Fast-Clear linker to actinium-225, a potent alpha-emitting radioisotope. This targeted alpha therapy is designed to deliver cytotoxic alpha radiation specifically to tumor cells that express FGFR3, an oncogenic driver overexpressed in several solid tumors, notably bladder, head and neck, and ovarian cancers. Developed for precision oncology, FPI-1966 aims to selectively irradiate and eradicate cancer cells while sparing normal tissue. Fusion Pharmaceuticals is the main developer, with ongoing Phase 1/2 clinical trials to evaluate safety, tolerability, dosimetry, pharmacokinetics, and preliminary efficacy in patients with advanced FGFR3-expressing solid tumors[1][2][3][4][7][10].
