Overview

Vofopitant is a small molecule drug that acts as a selective neurokinin 1 (NK1) receptor antagonist. It was developed as an antiemetic and investigated for its potential in treating conditions such as post-traumatic stress disorder (PTSD), primary insomnia, sleep initiation and maintenance disorders, social phobia, bipolar disorder, and nausea/vomiting. Despite showing anxiolytic effects in animal models and potent antiemetic activity similar to other NK1 antagonists, clinical trials did not demonstrate sufficient efficacy for marketing approval. The drug reached phase II clinical trials for PTSD and insomnia but development was discontinued due to lack of effectiveness[1][4][5].
