Drug intelligence / Profile preview
REMD-477 + insulin (volagidemab + insulin) is an investigational combination therapy being developed for the treatment of type 1 diabetes. The regimen consists of volagidemab (formerly REMD-477 or AMG-477), a fully human IgG2 monoclonal antibody that acts as a competitive antagonist of the human glucagon receptor (GCGR), administered alongside standard background insulin therapy. By blocking glucagon signaling, volagidemab inhibits hepatic glycogenolysis and gluconeogenesis, thereby lowering blood glucose levels and reducing exogenous insulin requirements. In clinical trials, the addition of volagidemab to insulin therapy has been shown to significantly improve glycemic control (as measured by HbA1c and continuous glucose monitoring parameters) and reduce daily insulin doses without increasing the risk of hypoglycemia.
