Overview
**Volasertib + nintedanib** is an experimental combination therapy for advanced solid tumors, evaluated in a phase I trial. **Volasertib** is a potent, selective small-molecule inhibitor of Polo-like kinase 1 (PLK1), a key regulator of mitosis, causing cell cycle arrest and apoptosis in cancer cells. **Nintedanib** is an oral, multi-targeted tyrosine kinase inhibitor of VEGFR, FGFR, and PDGFR (triple angiokinase inhibitor), acting as an anti-angiogenic agent. The combination aimed to improve antitumor activity by targeting both tumor cell division and the tumor microenvironment. In clinical studies, the regimen demonstrated a manageable safety profile with preliminary antitumor activity, including stable disease, partial responses, and a complete response in advanced cancer patients who failed prior therapies[1][2][5].
