Volrustomig + carboplatin + pemetrexed is a combination regimen under investigation for the treatment of several cancers, most notably metastatic non-small cell lung cancer and unresectable pleural mesothelioma. Volrustomig is a bispecific monoclonal antibody targeting both programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), thereby enhancing anti-tumor immunity by releasing two critical immune checkpoints. Carboplatin is a platinum-based chemotherapeutic agent that forms DNA adducts and induces apoptosis in rapidly dividing cells. Pemetrexed is a multi-targeted antifolate that inhibits several enzymes involved in purine and pyrimidine synthesis, leading to inhibition of DNA replication in cancer cells. The combination exploits both immunotherapeutic and cytotoxic mechanisms. The regimen is primarily delivered intravenously and has reached phase III clinical trial status for metastatic non-small cell lung cancer, with additional activity in pleural mesothelioma and other solid tumors[3][4][7][9].
