**Volrustomig + rilvegostomig** is an investigational combination of two bispecific monoclonal antibodies developed for cancer immunotherapy. **Volrustomig** is a bispecific antibody targeting both the programmed cell death protein 1 (*PD-1*) and cytotoxic T-lymphocyte-associated protein 4 (*CTLA-4*) immune checkpoints. It functions by simultaneously blocking PD-1 and CTLA-4 pathways, aiming to enhance T-cell activation and antitumor immunity. **Rilvegostomig** is a bispecific antibody targeting PD-1 and T-cell immunoreceptor with Ig and ITIM domains (*TIGIT*), inhibiting both checkpoints to further promote immune activation by preventing tumor-induced T-cell exhaustion and immune evasion. Both agents are administered intravenously and are being developed primarily for advanced solid tumors, including gastric cancer and hepatocellular carcinoma. The combination is being studied in early-phase (Phase 0 and Phase 2) clinical trials to evaluate synergistic immune activation and improved clinical outcomes in various cancer types[5][9][15][17][18].
