Overview

Vonifimod (NXC736, also known as SLB736) is a novel small-molecule drug developed by Nextgen Bioscience. It acts as a selective functional antagonist of the sphingosine 1-phosphate receptors S1P1 and S1P4, with immunomodulatory and anti-fibrotic effects. Vonifimod inhibits activation of the NLRP3 inflammasome and interferes with downstream inflammatory signaling pathways, including reducing IL-1β expression and blocking fibrotic processes via Smad 2/3/4 signaling. The drug is being investigated primarily for autoimmune diseases such as alopecia areata (in Phase 2 clinical trials), ulcerative colitis (Phase 1), and has also been studied preclinically for radiation-induced lung fibrosis and non-alcoholic steatohepatitis[1][2][3][5][6][7].
