vonoprazan

Vonoprazan is a first-in-class potassium-competitive acid blocker (PCAB) used to treat acid-related gastrointestinal disorders. It works by reversibly inhibiting the H+, K+-ATPase enzyme system (the gastric proton pump) in gastric parietal cells, thereby suppressing both basal and stimulated gastric acid secretion. Unlike traditional proton pump inhibitors (PPIs), which form covalent bonds with the enzyme, vonoprazan competitively blocks potassium binding at the active site of H+, K+-ATPase. This results in rapid and potent suppression of stomach acid production[3][6][8]. Vonoprazan is indicated for healing and maintenance of erosive esophagitis, relief of heartburn associated with erosive esophagitis, treatment of non-erosive gastroesophageal reflux disease (GERD), and as part of combination therapy for Helicobacter pylori eradication[1][2][7]. The drug was originally developed by Takeda.
