D-V14 is a 14-day dual therapy regimen investigated by the National Taiwan University Hospital for the first-line treatment of *Helicobacter pylori* infection. The regimen consists of vonoprazan, a potassium-competitive acid blocker (P-CAB), administered at 20 mg twice daily, and amoxicillin, a beta-lactam antibiotic, administered at 750 mg every 8 hours. Vonoprazan provides rapid, potent, and sustained suppression of gastric acid by competitively inhibiting the potassium-binding site of the gastric H+,K+-ATPase. This high level of acid suppression is critical for increasing the stability and efficacy of amoxicillin, which is highly pH-dependent, thereby improving eradication rates of *H. pylori* compared to traditional proton pump inhibitor (PPI)-based therapies.
