vonoprazan + minocycline is an investigational combination therapeutic regimen being evaluated for the eradication of Helicobacter pylori infection, particularly in patients with antibiotic-resistant strains. The regimen consists of vonoprazan, a first-in-class potassium-competitive acid blocker (P-CAB), and minocycline, a second-generation tetracycline antibiotic. Vonoprazan provides rapid, potent, and sustained inhibition of the gastric H+,K+-ATPase enzyme, maintaining a high intragastric pH that is essential for the stability and antimicrobial activity of antibiotics. Minocycline exerts its antibacterial effect by binding to the 30S ribosomal subunit, thereby inhibiting bacterial protein synthesis. This combination is primarily being researched by investigators at Tongji Hospital (Huazhong University of Science and Technology) as a high-efficacy dual therapy alternative to traditional bismuth-containing quadruple therapies or clarithromycin-based triple therapies.
