vonoprazan + pantoprazole

This is a combination of two acid-suppressing drugs, vonoprazan and pantoprazole. Vonoprazan is a potassium-competitive acid blocker (PCAB) that inhibits gastric acid secretion by competitively blocking the potassium-binding site of the H+,K+-ATPase enzyme in gastric parietal cells. Pantoprazole is a proton pump inhibitor (PPI) that irreversibly binds to and inhibits the same H+,K+-ATPase enzyme, but through covalent modification at cysteine residues. Both drugs reduce gastric acidity but via distinct mechanisms—vonoprazan acts as a reversible competitive inhibitor while pantoprazole forms an irreversible bond with the proton pump. This combination would theoretically provide potent and possibly complementary suppression of stomach acid production, though there are no standard clinical products or guidelines recommending their combined use together in one formulation or regimen[6][5][9].
