Vonoprazan + tetracycline is a combination therapy primarily investigated for the eradication of Helicobacter pylori infection. Vonoprazan, a potassium-competitive acid blocker (P-CAB), provides rapid, potent, and sustained suppression of gastric acid by inhibiting the H+, K+-ATPase enzyme in a reversible and K+-competitive manner. This acid suppression is crucial for enhancing the stability and efficacy of co-administered antibiotics. Tetracycline is a broad-spectrum bacteriostatic antibiotic that inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, preventing the attachment of aminoacyl-tRNA. The First Affiliated Hospital of Nanchang University has been a key developer in evaluating this combination, often as part of a bismuth-containing quadruple therapy (BQT) or modified regimens, to address the increasing global challenge of antibiotic-resistant H. pylori strains.
