This investigational triple therapy combination consists of vonoprazan, tripotassium dicitrate bismuthate, and amoxicillin, and is being studied by Kaohsiung Medical University for the treatment of Helicobacter pylori infection. Vonoprazan is a potassium-competitive acid blocker (P-CAB) that provides rapid and sustained inhibition of the gastric H+-K+ ATPase, creating an optimal pH environment for antibiotic activity. Tripotassium dicitrate bismuthate is a bismuth salt that exerts direct antimicrobial effects against H. pylori, prevents bacterial adhesion to the gastric mucosa, and enhances the efficacy of co-administered antibiotics. Amoxicillin is a beta-lactam antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins. This regimen is designed as a simplified alternative to traditional bismuth-based quadruple therapies, aiming to maintain high eradication rates with a reduced antibiotic burden.
