Overview

**Vopikitug (RG6292, RO7296682)** is an afucosylated human IgG1 monoclonal antibody developed by Roche that targets **CD25 (IL-2 receptor alpha subunit)** on regulatory T cells (Tregs). Engineered to selectively deplete Tregs via enhanced antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis while preserving IL-2 signaling on effector T cells, it aims to enhance antitumor immunity without broad immunosuppression. Administered intravenously, it demonstrated dose-dependent peripheral and intratumoral Treg depletion in phase 1 trials for advanced solid tumors, both as monotherapy and in combination with atezolizumab, but showed insufficient clinical efficacy leading to trial terminations. Preclinical data also supported potential in acute myeloid leukemia (AML), particularly in CD25-expressing leukemic stem cells.
