Vopimetostat + daraxonrasib is an investigational combination therapy co-developed by Tango Therapeutics and Revolution Medicines. Vopimetostat (TNG908) is an oral, once-daily, small molecule MTA-cooperative PRMT5 inhibitor that selectively targets cancer cells harboring MTAP (methylthioadenosine phosphorylase) deletions. Daraxonrasib (RMC-6236) is a RAS(ON) multi-selective inhibitor that targets oncogenic RAS mutations. The combination is designed to synergistically inhibit the RAS pathway and PRMT5 activity in MTAP-deleted, RAS-mutant tumors, such as pancreatic ductal adenocarcinoma (PDAC) and non-small cell lung cancer (NSCLC). Clinical data from Phase 1/2 trials have demonstrated high objective response rates and favorable progression-free survival in patients with metastatic PDAC.
