vorinostat + rituximab is a combination therapy consisting of the histone deacetylase (HDAC) inhibitor vorinostat and the CD20-directed monoclonal antibody rituximab. Vorinostat is a small molecule that inhibits the enzymatic activity of HDACs, leading to the accumulation of acetylated histones and other proteins, which induces cell cycle arrest and apoptosis in cancer cells. Rituximab is a chimeric monoclonal antibody that targets the CD20 antigen on the surface of B-lymphocytes, mediating cell lysis through complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC). This combination has been investigated by the City of Hope Medical Center in Phase II clinical trials for the treatment of indolent B-cell non-Hodgkin lymphomas, including follicular, marginal zone, and mantle cell lymphomas, to evaluate if the epigenetic modulation by vorinostat can enhance the efficacy of rituximab-based therapy.
