Voyager Therapeutics' APOE gene therapy is an investigational AAV-based gene therapy program for Alzheimer's disease. Currently in the IND-enabling stage, the therapy utilizes Voyager's proprietary TRACER-derived capsid technology for intravenous delivery across the blood-brain barrier. The therapy delivers a bifunctional payload designed to reduce the expression of the high-risk APOE4 variant while simultaneously delivering the protective APOE2 variant. Preclinical studies in mouse models have shown that this approach significantly reduces APOE4 levels in relevant brain regions while increasing APOE2 expression, potentially offering a disease-modifying treatment for carriers of the APOE4 allele.
