Drug intelligence / Profile preview
VP1-targeting small molecule is a first-in-class, orally available small-molecule antiviral candidate being developed by Orthogon Therapeutics (an affiliate of Pledge Therapeutics) for the treatment and prophylaxis of BK polyomavirus (BKPyV) and JC polyomavirus (JCPyV) infections, particularly in immunocompromised transplant recipients. The drug works by binding with high affinity to a highly conserved intracellular site on the viral capsid protein VP1, distinct from the variable antibody-binding cleft. This binding alters capsid architecture and blocks viral replication, providing pan-genotypic activity across BKPyV subtypes and JCPyV. The candidate is designed to penetrate renal sanctuaries of viral replication and maintain stability in urine, offering a high genetic barrier to resistance and avoiding the limitations of monoclonal antibodies.
