Drug intelligence / Profile preview
VP1-targeting small molecule inhibitor is a first-in-class, orally available small molecule antiviral candidate being developed by Orthogon Therapeutics (an affiliate of Pledge Therapeutics) to address BK polyomavirus (BKPyV) and JC polyomavirus (JCPyV) infections. The drug candidate targets the highly conserved intracellular domain of the viral capsid protein VP1, disrupting the viral lifecycle and capsid architecture. This mechanism provides pan-genotypic activity across BKPyV variants and related polyomaviruses, bypassing the limitations of neutralizing monoclonal antibodies that are prone to viral escape due to sequence variation in the outer BC-loop. It is designed to penetrate renal sanctuaries of viral replication and is being developed for the prevention and treatment of BKPyV reactivation, viruria, viremia, nephropathy, and hemorrhagic cystitis in kidney and hematopoietic transplant recipients.
