Overview

VP3.15 is a small molecule, non-ATP competitive inhibitor of glycogen synthase kinase 3 (GSK3), with activity against both alpha and beta isoforms. Developed by the Spanish National Research Council (CSIC), it belongs to the thiadiazolidinedione (TDZD) family of compounds. VP3.15 has been investigated for its potential to treat neurodegenerative and inflammatory conditions, such as Alzheimer's disease and multiple sclerosis, due to its ability to cross the blood-brain barrier and modulate neuroinflammation. Recent research also highlights its role in diabetic cardiomyopathy, where it inhibits a GSK3/NF-κB/REDD1 feed-forward loop that drives pro-inflammatory signaling and cardiac dysfunction.
