VP4896 is a proprietary, small, biodegradable implant developed by Voyager Pharmaceutical Corporation. It contains leuprolide acetate and a polymer. It was designed as a potential treatment for mild-to-moderate Alzheimer's disease based on the hypothesis that luteinizing hormone may contribute to disease development. VP4896 works as a gonadotropin-releasing hormone (GnRH) agonist that binds to and inactivates GnRH receptors on the pituitary gland, aiming to decrease the amount of luteinizing hormone released. Voyager Pharmaceutical Corporation conducted Phase III clinical trials for VP4896, including the ALADDIN Study, to assess its safety and efficacy in treating subjects with mild-to-moderate Alzheimer's disease. Primary efficacy endpoints included changes in ADAS-Cog and ADCS-CGIC scores. Pooled analysis from two studies reportedly showed significant improvement after 48 weeks.
