Overview

VQ-201 is an allosteric small molecule antagonist of the complement C5a receptor 1 (C5aR1), developed by Vanqua Bio for the treatment of inflammatory disorders. It targets complement-mediated inflammation by robustly blocking C5a-mediated chemotaxis and activation of pathogenic immune cells in vitro, and has demonstrated protection of renal function in a mouse model of autoantibody-driven renal diseases in vivo. VQ-201 is designed to avoid the structural liabilities associated with first-generation small molecule C5aR1 antagonists, which have been linked to idiosyncratic hepatotoxicity. The drug has shown a promising safety and tolerability profile in preclinical studies and is projected for once-daily oral administration in humans. Vanqua Bio plans to initiate Phase 1 clinical trials for VQ-201 in early 2026, with potential indications including peripheral inflammatory disorders such as kidney diseases, as well as central nervous system conditions like Alzheimer's disease and multiple sclerosis[1][3][2][5].
