Overview

VR147 is an investigational oncolytic immunotherapy based on a modified Herpes Simplex Virus type 1 (HSV-1) platform, developed by Virogin Biotech. It is engineered using the company's proprietary "Synerlytic" platform, which incorporates dual transcriptional and translational regulation to ensure high tumor selectivity and safety. The virus is attenuated through the deletion of the neurovirulence factor ICP34.5 and is designed to replicate preferentially in malignant cells by exploiting dysregulated signaling pathways, such as the eIF4E translational machinery. VR147 functions by inducing direct oncolysis and promoting immunogenic cell death, which facilitates the release of tumor-associated antigens and stimulates a systemic anti-tumor immune response. It serves as the foundational backbone for Virogin's clinical-stage candidates, such as VG161, which are further modified to express therapeutic payloads like cytokines and checkpoint inhibitors.
