VR496 is an inhaled formulation of unfractionated heparin developed by Vectura for the treatment of chronic respiratory conditions, including cystic fibrosis, asthma, and chronic obstructive pulmonary disease (COPD). While heparin is traditionally known as a systemic anticoagulant, VR496 utilizes its non-anticoagulant properties to act as a multi-modal therapeutic agent. Its mechanism of action involves the inhibition of neutrophil elastase, a protease that contributes to airway wall degradation and mucus hypersecretion, as well as acting as a mucolytic by reducing the viscosity of sputum. Additionally, it exhibits anti-inflammatory properties by interfering with the recruitment of inflammatory cells. Development reached Phase 2 clinical trials for cystic fibrosis before being discontinued.
