Overview

VRC-ADJDNA004-IL2-VP is a plasmid DNA adjuvant developed by the Vaccine Research Center (VRC) of the National Institute of Allergy and Infectious Diseases (NIAID). It encodes a fusion protein consisting of human interleukin-2 (IL-2) and the Fc portion of human immunoglobulin G (Ig). This genetic adjuvant is designed to be co-administered with DNA vaccines, such as the HIV-1 vaccine VRC-HIVDNA009-00-VP, to enhance their immunogenicity. The expressed IL-2/Ig fusion protein acts as a potent cytokine that stimulates the proliferation and activation of T cells and natural killer (NK) cells, thereby boosting the cellular immune response against the vaccine antigens. The inclusion of the Ig Fc fragment is intended to improve the stability and extend the half-life of the IL-2 protein in vivo. It has been evaluated in Phase 1 clinical trials for safety and its ability to modulate immune responses in healthy, HIV-uninfected adults.
