VRC-HIVADV054-00-VP (rAd5 gag-pol) is a recombinant adenovirus serotype 5 (rAd5) vector-based vaccine developed by the Vaccine Research Center (VRC) at the National Institute of Allergy and Infectious Diseases (NIAID). The vaccine is engineered to express an HIV-1 clade B Gag-Pol fusion protein, targeting the internal structural and enzymatic proteins of the virus. It is designed to elicit both cellular and humoral immune responses to prevent or control HIV-1 infection. VRC-HIVADV054-00-VP has been evaluated in Phase 1 clinical trials, including HVTN 085, which explored polytopic administration strategies—such as single-site versus divided-dose delivery—to optimize the immunogenicity of the VRC rAd5 HIV vaccine platform.
