Overview

VRC-HIVDNA016-00-VP is a multiclade HIV-1 DNA plasmid vaccine developed by the Vaccine Research Center (VRC) of the National Institute of Allergy and Infectious Diseases (NIAID). The vaccine is composed of six DNA plasmids in equal concentrations, encoding HIV-1 Gag, Pol, and Nef proteins from clade B, and Envelope (Env) glycoproteins from clades A, B, and C. It was primarily evaluated as a priming component in heterologous prime-boost regimens, often paired with the recombinant adenoviral vector vaccine VRC-HIVADV014-00-VP. The goal of the vaccine was to elicit broad cellular and humoral immune responses against diverse HIV-1 strains. However, the pivotal Phase 2b HVTN 505 trial was halted in 2013 after an interim analysis demonstrated that the vaccine regimen did not prevent HIV infection or reduce viral load in those who became infected.
