VRC-HIVDNA044-00-VP is an experimental multiclade DNA vaccine developed by the Vaccine Research Center (VRC) of the National Institute of Allergy and Infectious Diseases (NIAID). The vaccine is designed to prevent HIV-1 infection by eliciting broad cellular (T-cell) and humoral (B-cell) immune responses. It typically consists of six distinct mammalian expression plasmids encoding various HIV-1 antigens: a Gag-Pol-Nef fusion protein and Envelope (Env) glycoproteins from clades A, B, and C. In clinical development, VRC-HIVDNA044-00-VP has been evaluated primarily as a priming agent in heterologous prime-boost regimens, followed by boosting with viral vector vaccines such as recombinant adenovirus serotype 5 (rAd5) or serotype 35 (rAd35) to enhance the magnitude and breadth of the immune response.
