VRC-WNVDNA017-00-VP is a DNA vaccine candidate developed for the prevention of West Nile virus (WNV) infection. It consists of a single, closed, circular plasmid DNA macromolecule (VCL-6428) designed to express the prM and E proteins of the WNV envelope glycoprotein. The construct includes a modified signal sequence from Japanese encephalitis virus (JEV) fused upstream of WNV prM and E coding sequences, cloned into the VR-1012 expression vector with a cytomegalovirus (CMV) promoter backbone. Upon in vitro expression, it forms noninfectious subviral particles (SVPs), mimicking viral antigens to elicit an immune response without risk of causing infection. The vaccine is incapable of replication in animal cells and cannot generate infectious virions even if recombination occurs. It was produced under current good manufacturing practice conditions by Vical using bacterial fermentation and purified for intramuscular injection[2][1][3].
