Overview

VRC-WNVDNA020-00-VP is a second-generation DNA vaccine candidate developed for the prevention of West Nile Virus (WNV) infection. The vaccine consists of a single, closed, circular plasmid DNA macromolecule (VCL-8111) engineered to express the prM and E proteins of the WNV envelope glycoprotein. These sequences are derived from the NY99 human WNV isolate. The plasmid uses a modified CMV/R promoter, which includes regulatory elements from HTLV-1 to enhance transcription and posttranscriptional events, resulting in improved protein expression and immunogenicity compared to earlier constructs. Upon administration, in vitro expression leads to formation of noninfectious subviral particles (SVPs), stimulating an immune response without risk of generating infectious virions. The vaccine was produced under Good Manufacturing Practices by bacterial fermentation with kanamycin selection[1]. Early clinical trials demonstrated that it induces neutralizing antibodies against WNV in healthy adults[3].
