VRC01.23LS is a broadly neutralizing human monoclonal antibody (bnAb) engineered to target the CD4 binding site (CD4bs) of the HIV-1 envelope glycoprotein gp120[4][6]. It is designed for enhanced potency and extended half-life compared to earlier antibodies in its class, such as VRC01, through LS mutations that improve binding to the neonatal Fc receptor and thus prolong serum persistence[6]. The primary mechanism of action is direct neutralization of HIV-1 by blocking viral entry into host cells via competitive inhibition at the CD4bs. Developed as part of efforts to prevent or treat HIV infection, it has been evaluated in first-in-human clinical trials for safety, tolerability, pharmacokinetics, and immunogenicity in healthy adults[1][4][6].
