Overview

VRC5288 (code name VRC-ZKADNA085-00-VP) is an investigational DNA vaccine developed by the Vaccine Research Center (VRC) at the National Institute of Allergy and Infectious Diseases (NIAID). It consists of a single closed-circular DNA plasmid encoding a chimeric precursor membrane (prM) and envelope (E) protein derived from both Zika virus and Japanese encephalitis virus. The vaccine is designed to induce immune responses against Zika virus by expressing these structural proteins in host cells, leading to the production of neutralizing antibodies. Preclinical studies demonstrated strong immunogenicity and protection in animal models. In phase 1 clinical trials, it was administered intramuscularly to healthy adults to assess safety and immunogenicity for prevention of Zika virus infection[1][2][5][8].
