Overview

VRN04 is a small molecule inhibitor of Receptor-interacting serine/threonine-protein kinase 1 (RIPK1) being developed by Voronoi Productions for the treatment of autoimmune and inflammatory diseases. RIPK1 is a critical signaling node that regulates cell fate (apoptosis and necroptosis) and the production of inflammatory cytokines in response to various stimuli, including TNF-alpha. By selectively inhibiting the kinase activity of RIPK1, VRN04 is designed to block necroptotic cell death and reduce systemic inflammation without interfering with pro-survival signaling. The candidate is currently undergoing Phase 1/2 clinical evaluation, representing a targeted approach to managing chronic inflammatory conditions through the regulation of programmed necrosis.
