VRON-0200-AdC6 is a first-in-class therapeutic immunotherapy designed for the functional cure of chronic hepatitis B virus (HBV) infection. It utilizes a chimpanzee adenoviral vector serotype 6 (AdC6) to deliver genetically encoded HBV core and polymerase antigens along with a checkpoint modifier, glycoprotein D. The therapy aims to eliminate cccDNA-infected hepatocytes and stimulate expansion of new HBV-specific CD8+ T cells by overcoming T cell exhaustion through checkpoint modification. This approach broadens and amplifies the immune response against HBV by targeting subdominant epitopes not activated during natural infection, thereby restoring immune control over the virus. VRON-0200 is administered intramuscularly and has demonstrated safety, tolerability, immunogenicity, and anti-HBV activity in Phase 1b clinical trials[1][2][3][4][5][8]. Developed as an interferon-sparing option for chronic HBV patients on nucleos(t)ide antiviral therapy, it does not directly target HBsAg but has shown declines in surface antigen levels in some patients[5][8].
