VRP-BORIS (also known as VRP-mBORIS) is a therapeutic cancer vaccine consisting of a recombinant Venezuelan Equine Encephalitis (VEE) virus-like replicon particle (VRP) vector that expresses a modified version of the cancer-testis antigen Brother of Regulator of Imprinted Sites (BORIS, also known as CTCFL). The modified BORIS antigen lacks the DNA-binding zinc finger domain to prevent potential oncogenic activity and off-target autoimmune responses. VRP-BORIS is designed to target dendritic cells in vivo, promoting antigen presentation and eliciting a robust cellular immune response (including T-helper cell proliferation and interferon-gamma secretion) against BORIS-expressing cancer cells, particularly cancer stem cells. It has been investigated in preclinical models of breast cancer.
