VRTX153 is a small molecule inhibitor targeting the KRAS G12D mutation, which is a common oncogenic driver in several solid tumors, most notably pancreatic cancer. The drug acts by inhibiting the GTPase activity of mutant KRAS G12D protein, thereby disrupting downstream signaling pathways that promote tumor growth and survival. VRTX153 is being developed by Vrise Therapeutics and is currently in preclinical development. Its primary indication is pancreatic cancer, but it may also have potential applications in other cancers harboring the KRAS G12D mutation[1][2][3][7].
