VRX1023

VRX1023 is a small molecule potassium channel opener that specifically targets the KCNQ2/3 (Kv7.2/7.3) channels. Developed by Valeant Pharmaceuticals (now Bausch Health), it was designed as a second-generation follow-up to retigabine (ezogabine) with the aim of improving potency and selectivity for the treatment of epilepsy and neuropathic pain. By facilitating the opening of KCNQ channels, VRX1023 stabilizes the neuronal membrane potential and reduces hyperexcitability, which is a hallmark of seizure activity and chronic pain states. Development of the compound was discontinued during early clinical stages, likely due to shifting strategic priorities and the safety concerns, such as skin and retinal pigmentation, that emerged with the first-generation compound retigabine.
