Overview

VS-041 is an orally bioavailable small molecule drug developed by Vasa Therapeutics for the treatment of heart failure with preserved ejection fraction (HFpEF), a form of heart failure characterized by diastolic dysfunction and cardiac fibrosis. It acts as a selective inhibitor of matrix metalloproteinases (MMP2, MMP9, and MMP13), targeting collagen remodeling in the myocardium. By inhibiting the release of endotrophin—a signaling collagen fragment implicated in fibrotic and inflammatory responses—VS-041 aims to reduce cardiac fibrosis and improve diastolic function. Preclinical studies have shown robust efficacy in animal models, with significant improvements in diastolic function and reductions in left ventricular fibrosis without affecting blood pressure or ejection fraction. The drug is currently undergoing Phase 1 clinical trials to assess safety, tolerability, and pharmacokinetics[1][2][4][5][6][7].
