VS-214 is a long-acting apelin analog being developed as a peptide therapeutic for the treatment of peripheral arterial disease (PAD) and various rare neuromuscular diseases. Developed by Vasa Therapeutics, it acts as an agonist of the apelin receptor, a G protein-coupled receptor that modulates cardiovascular and musculoskeletal signaling pathways. Preclinical evidence suggests that VS-214 can increase muscle strength and endurance, preserve muscle function, and enhance cardiac output. Its targeted indications include facioscapulohumeral muscular dystrophy (FSHD), spinal muscular atrophy (SMA), spinal cord injury, and sarcopenia. The drug is currently in preclinical development, with clinical trials expected to commence in 2026.
