Overview

This is a combination therapy consisting of three agents: VS-4718, nab-paclitaxel, and gemcitabine. VS-4718 is an orally bioavailable small molecule inhibitor of focal adhesion kinase (FAK), which blocks FAK autophosphorylation at Tyr397 and disrupts integrin-mediated signaling pathways such as ERK, JNK/MAPK, and PI3K/Akt. This inhibition reduces cancer stem cell populations and impairs tumor cell migration, proliferation, and survival[1][3][4]. Nab-paclitaxel (nanoparticle albumin-bound paclitaxel) is a microtubule inhibitor that interferes with mitosis in rapidly dividing cells; it is marketed under the brand name Abraxane[7]. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA strands during replication; it is marketed as Gemzar[7]. The combination of these drugs has been investigated for synergistic antitumor effects in various cancers including pancreatic cancer.
