Overview

The VSIG4 monoclonal antibody is a preclinical-stage therapeutic program being developed by Pear Bio for the treatment of lung, ovarian, and liver cancers. The antibody targets V-set immunoglobulin domain-containing 4 (VSIG4), an immunosuppressive receptor predominantly expressed on tumor-associated macrophages (TAMs) that functions as a negative regulator of T cell activation. By blocking the interaction between VSIG4 and its ligands, the antibody is designed to alleviate T cell suppression and repolarize TAMs from an M2-like immunosuppressive state to a pro-inflammatory phenotype, thereby stimulating a robust anti-tumor immune response. Pear Bio has reported that multiple hit antibodies in this program have demonstrated a greater than 100-fold improvement in binding affinity (KD) compared to known antibodies and target diverse domains and epitopes of the VSIG4 protein.
