VSR-1045 is an IgG1 monoclonal antibody designed to target a disease-specific fibronectin (Fn) mechanoswitch for the treatment of idiopathic pulmonary fibrosis (IPF). Developed by Vasarya Therapeutics in collaboration with the University of Virginia, the antibody specifically binds to a novel integrin-binding site on fibronectin that is exposed only under mechanical force, a condition characteristic of the aberrant extracellular matrix deposition in fibrotic lung tissue. By targeting this mechanoswitch, VSR-1045 aims to inhibit profibrotic signaling pathways, including TGFβ-induced SMAD2 nuclear translocation and YAP1 activation, thereby reducing fibroblast activation and promoting regenerative gene signatures. Preclinical studies in bleomycin-induced lung fibrosis models have demonstrated its potential to reduce fibrosis scores and restore normal alveolar structures.
