Overview

**VSV-CD133** is a genetically engineered **oncolytic virus** based on vesicular stomatitis virus (VSV), in which the viral glycoprotein has been replaced by measles virus (MV) glycoproteins that have been further modified to target the cell surface marker **CD133 (prominin-1)** using a CD133-specific single-chain antibody fragment (scFv). This engineering confers selective infection and killing of **CD133-positive cancer stem cells**, which are implicated in tumor resistance, recurrence, and metastasis. VSV-CD133 demonstrates highly potent oncolytic activity against CD133-positive tumor models, including glioma and hepatocellular carcinoma, significantly inhibiting tumor growth and prolonging survival in preclinical animal models when administered intratumorally or intravenously. However, fatal neurotoxicity has been observed in some brain tumor models, indicating the need for careful preclinical safety evaluation. VSV-CD133 is an example of a targeted **oncolytic virus therapy**, representing a novel, receptor-targeted approach in cancer virotherapy[2][3][4].
