Overview

VSV-GP128 is a recombinant, live-attenuated vesicular stomatitis virus (VSV) genetically modified to express the envelope glycoprotein (GP) of a non-neurotropic strain of lymphocytic choriomeningitis virus (LCMV), specifically the WE-HPI strain, instead of its native glycoprotein. It is being developed as an oncolytic viral vaccine and immunostimulant for use in heterologous prime-boost regimens with ATP128, primarily targeting stage IV colorectal cancer. The approach aims to induce strong tumor antigen-specific cytotoxic T cell responses and enhance anti-tumor immunity, especially when combined with immune checkpoint inhibitors such as BI 754091 (ezabenlimab). VSV-GP128 is administered intravenously as part of clinical trials evaluating its safety, tolerability, and immunogenicity in patients with microsatellite stable/mismatch repair-proficient colorectal cancer[2][4][5].
