**VSV-hgp100** is an engineered oncolytic virus based on vesicular stomatitis virus (VSV) that expresses the human melanoma-associated antigen glycoprotein 100 (gp100). Its mechanism combines direct oncolysis with immune-mediated anti-tumor activity: VSV infects and kills tumor cells and, by delivering the gp100 antigen, primes and activates gp100-specific T cells (primarily CD8^+^ cytotoxic T lymphocytes) to target and kill tumor cells[1][2][3][5][7]. VSV-hgp100 enhances antitumor immunity synergistically when combined with adoptive cell transfer (e.g., pmel-1 T cells), checkpoint inhibitors (such as anti-PD-L1 or anti-CTLA-4 therapies), or cancer vaccines[1][2][3][7]. In preclinical mouse models of melanoma, systemic VSV-hgp100 administration led to regression of established tumors and long-term, recurrence-free survival in many cases[1][3]. The virus acts as both an oncolytic agent and a cancer vaccine/adjuvant, stimulating both innate and adaptive immunity against tumor-associated antigens[2][5][7].
